Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002439.5(MSH3):c.89C>G (p.Ser30Cys), citing Ambry Variant Classification Scheme 2023: The p.S30C variant (also known as c.89C>G), located in coding exon 1 of the MSH3 gene, results from a C to G substitution at nucleotide position 89. The serine at codon 30 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
