Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.2051G>C (p.Arg684Pro), citing Ambry Variant Classification Scheme 2023: The p.R684P variant (also known as c.2051G>C), located in coding exon 19 of the PLOD1 gene, results from a G to C substitution at nucleotide position 2051. The arginine at codon 684 is replaced by proline, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
