NM_005902.4(SMAD3):c.721A>C (p.Asn241His) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N241H variant (also known as c.721A>C), located in coding exon 6 of the SMAD3 gene, results from an A to C substitution at nucleotide position 721. The asparagine at codon 241 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
