NM_198253.3(TERT):c.2181G>T (p.Glu727Asp) was classified as Uncertain significance for Dyskeratosis congenita by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E727D variant (also known as c.2181G>T), located in coding exon 6 of the TERT gene, results from a G to T substitution at nucleotide position 2181. The glutamic acid at codon 727 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
