Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.2733G>C (p.Gln911His), citing Ambry Variant Classification Scheme 2023: The p.Q911H variant (also known as c.2733G>C), located in coding exon 24 of the POLE gene, results from a G to C substitution at nucleotide position 2733. The glutamine at codon 911 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
