Uncertain significance for Syndromic multisystem autoimmune disease due to ITCH deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031483.7(ITCH):c.638C>T (p.Pro213Leu), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 213 of the ITCH protein (p.Pro213Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ITCH-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
