Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001031679.3(MSRB3):c.-118C>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with isoleucine at codon 11 of the MSRB3 protein (p.Leu11Ile). The leucine residue is weakly conserved and there is a small physicochemical difference between leucine and isoleucine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with MSRB3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
