Likely pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_182961.4(SYNE1):c.16155C>G (p.Tyr5385Ter): DNA sequence analysis of the SYNE1 gene demonstrated a sequence change, c.15942C>G, that results in the creation of a premature stop codon at amino acid position 5314, p.Tyr5314*. This sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated SYNE1 protein with potentially abnormal function. This sequence change does not appear to have been previously described in individuals with SYNE1-related disorders and has also not been described in population databases such as ExAC and gnomAD. Based on these evidences, this sequence change is classified as likely pathogenic.
