Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_002474.3(MYH11):c.2226C>A (p.Asp742Glu), citing Ambry Variant Classification Scheme 2023: The p.D742E variant (also known as c.2226C>A), located in coding exon 17 of the MYH11 gene, results from a C to A substitution at nucleotide position 2226. The aspartic acid at codon 742 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
