Submitter rationale: CFI p.Asp208Asn (c.622G>A) is a missense variant that changes the amino acid at residue 208 from Aspartic acid to Asparagine. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:35619721). Functional studies have been reported; however, the significance of the findings remain unclear (PMID:35619721). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is not damaging. In conclusion, we classify CFI p.Asp208Asn (c.622G>A) as a variant of unknown significance.
