NM_172351.3(CD46):c.98-1G>C was classified as Pathogenic for Atypical hemolytic-uremic syndrome by Genomenon, Inc, Genomenon, Inc, citing Genomenon Sequence Variant Interpretation Standards: CD46 c.98-1G>C is a canonical splice variant located in the acceptor splice region in intron 1. It is predicted to affect mRNA splicing, leading to a deleterious effect on the CD46 protein. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:34169201;16621965). The variant was found to segregate with disease in at least one affected family (PMID:16621965). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify CD46 c.98-1G>C as a pathogenic variant.
