Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000016.9:g.(?_30194730)_(30200398_?)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the duplication of exons 1-11 (i.e. the full coding sequence) of the CORO1A gene. A presumed nomenclature of c.(?_-318)_(*113_?)dup has been designated for the purposes of this classification. It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). Since exact breakpoints of this duplication are not known, it might extend beyond the assayed region of the CORO1A gene, including other flanking genes. A similar duplication spanning CORO1A gene and many other genes was found at a frequency of 4.6e-05 in 21694 control chromosomes (gnomAD Structural Variants dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(?_-318)_(*113_?)dup in individuals affected with Severe Combined Immunodeficiency and no experimental evidence demonstrating its impact on protein function have been reported. One ClinVar submitter has assessed the variant since 2014: the variant was classified as of uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
