This variant lies in the MEFV gene (transcript NM_000243.3) at coding-DNA position 1883, where G is replaced by A; at the protein level this means replaces arginine at residue 628 with lysine — a missense variant. Submitter rationale: Variant summary: MEFV c.1883G>A (p.Arg628Lys) results in a conservative amino acid change located in the B30.2/SPRY domain (IPR001870) of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4.1e-06 in 241538 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1883G>A has been reported in the INFEVERS database in at least one individual affected with Familial Mediterranean Fever, however, this report does not provide unequivocal conclusions about association of the variant with Familial Mediterranean Fever. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
