Submitter rationale: The missense variant c.994C>T in STAT3 has been reported in heterozygous state in individuals affected with Hyper-IgE recurrent infection syndrome (Soltész B, Tóth B, Shabashova N, et al, 2013; Jiao, Hong et al., 2008). The p.His332Tyr variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. The amino acid change p.His332Tyr in STAT3 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid His at position 332 is changed to a Tyr changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Likely Pathogenic.
