This variant lies in the GJB6 gene (transcript NM_001110219.3) at coding-DNA position 593, where C is replaced by A; at the protein level this means replaces alanine at residue 198 with glutamic acid — a missense variant. Submitter rationale: ClinVar contains an entry for this variant (Variation ID: 1699284). This sequence change replaces alanine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 198 of the GJB6 protein (p.Ala198Glu).
