Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015065.3(EXPH5):c.4917C>A (p.Cys1639Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Cys1639*) in the EXPH5 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 351 amino acid(s) of the EXPH5 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with epidermolysis bullosa simplex (PMID: 37462267; internal data). ClinVar contains an entry for this variant (Variation ID: 1698844). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
