This variant lies in the LRPPRC gene (transcript NM_133259.4) at coding-DNA position 124, where G is replaced by A; at the protein level this means replaces alanine at residue 42 with threonine — a missense variant. Submitter rationale: Variant summary: LRPPRC c.124G>A (p.Ala42Thr) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00012 in 57198 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.124G>A in individuals affected with Leigh Syndrome, French-Canadian Type and no experimental evidence demonstrating its impact on protein function have been reported.
