Likely pathogenic for Hermansky-Pudlak syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_022081.6(HPS4):c.1535C>G (p.Ser512Ter), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HPS4 c.1535C>G (p.Ser512X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic within ClinVar (e.g. c.1891C>T [p.Gln631Ter]; c.1818_1819insC [p.Tyr607fs]). The variant allele was found at a frequency of 8e-06 in 251482 control chromosomes (gnomAD). To our knowledge, no occurrence of c.1535C>G in individuals affected with Hermansky-Pudlak Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
