This variant lies in the VWF gene (transcript NM_000552.5) at coding-DNA position 5311, where G is replaced by A; at the protein level this means replaces glycine at residue 1771 with arginine — a missense variant. Submitter rationale: Variant summary: VWF c.5311G>A (p.Gly1771Arg) results in a non-conservative amino acid change located in the von Willebrand factor, type A domain (IPR002035) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site, and one predicts the variant abolishes a cryptic 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.2e-05 in 249874 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5311G>A has been reported in the literature in individuals affected with Von Willebrand Disease (Corrales_2012, Borras_2017).
