Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001145809.2(MYH14):c.488G>A (p.Arg163Gln), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the MYH14 gene (transcript NM_001145809.2) at coding-DNA position 488, where G is replaced by A; at the protein level this means replaces arginine at residue 163 with glutamine — a missense variant. Submitter rationale: Variant summary: MYH14 c.488G>A (p.Arg163Gln) results in a conservative amino acid change located in the Myosin head, motor domain (IPR001609) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 249274 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.488G>A in individuals affected with Deafness, Autosomal Dominant 4 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
