Pathogenic for FGFR2-related craniosynostosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000141.5(FGFR2):c.1977G>T (p.Lys659Asn), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects FGFR2 function (PMID: 17803937). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with FGFR2-related craniosynostosis (PMID: 11781872, 16418739, 28990276; Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 659 of the FGFR2 protein (p.Lys659Asn).
