This variant lies in the SMARCE1 gene (transcript NM_003079.5) at coding-DNA position 68, where C is replaced by G; at the protein level this means replaces proline at residue 23 with arginine — a missense variant. Submitter rationale: The p.P23R variant (also known as c.68C>G), located in coding exon 3 of the SMARCE1 gene, results from a C to G substitution at nucleotide position 68. The proline at codon 23 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Missense and in-frame variants in SMARCE1 are known to cause neurodevelopmental disorders; however, such associations with increased risk of meningiomas are exceedingly rare (Kosho T et al.
