This variant is absent from in the gnomAD v3.1.1 database suggesting it is not a common benign variant in the populations represented in this database. This variant creates a premature translational stop signal atexon 15 of 22 and is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ANO3 cause disease. Based on the available evidence, the nonsense variant c.702C>A, p.Cys234Ter in the ANO3 gene is classified as a Variant of Uncertain Significance.
