NM_000492.4(CFTR):c.3367+196C>T was classified as Benign by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFTR c.3367+196C>T is located at a position not widely known to affect splicing. The variant allele was found at a frequency of 0.14 in 12198 control chromosomes in the gnomAD database, including 127 homozygotes. The observed variant frequency is approximately 11 fold of the estimated maximal expected allele frequency for a pathogenic variant in CFTR causing Cystic Fibrosis phenotype (0.013), strongly suggesting that the variant is benign. To our knowledge, no occurrence of c.3367+196C>T in individuals affected with Cystic Fibrosis and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as benign.
