This variant lies in the KDM1A gene (transcript NM_001009999.3) at coding-DNA position 2314, where G is replaced by A; at the protein level this means replaces valine at residue 772 with methionine — a missense variant. Submitter rationale: Variant summary: AOF2 c.2314G>A (p.Val772Met) (aka. KDM1A c.2242G>A (p.V748M)) results in a conservative amino acid change located in the Amine oxidase domain (IPR002937) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250720 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2314G>A in individuals affected with Palatal Anomalies-Widely Spaced Teeth-Facial Dysmorphism-Developmental Delay Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
