This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 2415, where C is replaced by G; at the protein level this means replaces isoleucine at residue 805 with methionine — a missense variant. Submitter rationale: The IFIH1 c.2415C>G (p.Ile805Met) missense variant results in the substitution of isoleucine at position 805 with methionine. To our knowledge, this variant has not been reported in the peer-reviewed literature. This variant is not found in version 2.1.1 or version 3.1.2 of the Genome Aggregation Database. The c.2415C>G variant lies within one of two conserved core helicase domains. Rice et al. (2020) reported several de novo missense changes nearby to Ile805 and noted that reported variants tend to cluster near the ATP binding site within the helicase domain.
