NM_003995.4(NPR2):c.1579C>T (p.Leu527Phe) was classified as Pathogenic for Disproportionate short stature by Department of Endocrinology and Genetics, Fuzhou Children’s Hospital of Fujian Medical University: The proband, a 7.6-year-old girl, was admitted to hospital due to her short stature since early childhood. Her height and weight were 110.2cm (-3.06SDS) and 17.5kg (-0.33SDS), respectively. Her sitting height was 62.2cm, and sitting height/height ratio was 0.56 (2.35SDS). A spinal X-ray revealed slight lumbar scoliosis . The bone age was 5.3 years using the Tanner-Whitehouse 3 method. Her parents had proportionate bodies. WES and Sanger sequencing revealed a heterozygous NPR2 variant c.1579C>T,p.Leu527Phe in this patient. Neither parents carried this variant, suggesting that it had occurred de novo. Additionally, in vitro functional studies indicate that the Leu527Phe variant has 38.2% cGMP production capacity of wild-type, while the heterozygous variant WT/Leu527Phe retained 50.2% of activity VS. wild-type.
