Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_007194.4(CHEK2):c.592+5T>C, citing Sema4 Curation Guidelines. This variant lies in the CHEK2 gene (transcript NM_007194.4) at 5 bases into the intron immediately after coding-DNA position 592, where T is replaced by C. Submitter rationale: To the best of our knowledge, the CHEK2 c.592+5T>C (p.?) variant has not been reported in individuals with CHEK2-related disease. It was not observed in the large and broad cohorts of Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has not been reported in ClinVar. In silico tools suggest that the variant does not have an impact on splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
