Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_002528.7(NTHL1):c.749C>T (p.Ser250Phe), citing Sema4 Curation Guidelines. This variant lies in the NTHL1 gene (transcript NM_002528.7) at coding-DNA position 749, where C is replaced by T; at the protein level this means replaces serine at residue 250 with phenylalanine — a missense variant. Submitter rationale: The NTHL1 c.773C>T (p.S258F) variant has not been reported in the literature to our knowledge. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654), nor in ClinVar. Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
