NM_002439.5(MSH3):c.175G>C (p.Ala59Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the MSH3 gene (transcript NM_002439.5) at coding-DNA position 175, where G is replaced by C; at the protein level this means replaces alanine at residue 59 with proline — a missense variant. Submitter rationale: The MSH3 c.175G>C (p.A59P) variant has not been reported in the literature to our knowledge. It was observed in 2/61152 chromosomes of the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has not been reported in ClinVar. Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
