Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000551.4(VHL):c.464-1262G>A, citing Sema4 Curation Guidelines: The VHL c.464-1262G>A variant has not been reported in the literature to our knowledge. The variant is not observed in any sub-population with a sufficient number of total alleles (>=2000), but is observed in 2/30496 chromosomes across all populations in the large and broad cohorts in the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has not been reported in ClinVar. Algorithms developed to predict the effect of sequence changes on RNA splicing do not suggest negative effect, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
