Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4993C>G (p.Gln1665Glu), citing Ambry Variant Classification Scheme 2023: The p.Q1665E variant (also known as c.4993C>G), located in coding exon 38 of the TSC2 gene, results from a C to G substitution at nucleotide position 4993. The glutamine at codon 1665 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
