Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.50C>G (p.Pro17Arg), citing Ambry Variant Classification Scheme 2023: The p.P17R variant (also known as c.50C>G), located in coding exon 1 of the TSC1 gene, results from a C to G substitution at nucleotide position 50. The proline at codon 17 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
