Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.1931C>A (p.Ser644Tyr), citing Ambry Variant Classification Scheme 2023: The p.S644Y variant (also known as c.1931C>A), located in coding exon 17 of the NF1 gene, results from a C to A substitution at nucleotide position 1931. The serine at codon 644 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
