Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.1873T>C (p.Ser625Pro), citing Ambry Variant Classification Scheme 2023: The p.S625P variant (also known as c.1873T>C), located in coding exon 4 of the MSH6 gene, results from a T to C substitution at nucleotide position 1873. The serine at codon 625 is replaced by proline, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
