Uncertain significance for Fanconi anemia — the classification assigned by Sema4, Sema4 to NM_000135.4(FANCA):c.3018G>T (p.Leu1006Phe), citing Sema4 Curation Guidelines. This variant lies in the FANCA gene (transcript NM_000135.4) at coding-DNA position 3018, where G is replaced by T; at the protein level this means replaces leucine at residue 1006 with phenylalanine — a missense variant. Submitter rationale: The FANCA c.3018G>T (p.L1006F) variant has not been reported in the literature to our knowledge. It was observed in 1/113758 chromosomes of the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has not been reported in ClinVar. Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
