Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000059.4(BRCA2):c.-39-11C>T, citing Sema4 Curation Guidelines. This variant lies in the BRCA2 gene (transcript NM_000059.4) at 11 bases into the intron immediately before 39 bases upstream of the translation start (5' untranslated region), where C is replaced by T. Submitter rationale: The BRCA2 c.-39-11C>T variant has not been reported in the literature to our knowledge. It was observed in 2/34414 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has not been reported in ClinVar. In silico tools suggest that the variant does not impact splicing, though these predictions have not been confirmed by functional studies. There is no indication that this variant causes disease, but the evidence is insufficient currently to prove that conclusively. Thus, the clinical significance of this variant is currently uncertain.
