Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024529.5(CDC73):c.1006G>T (p.Ala336Ser), citing Ambry Variant Classification Scheme 2023: The p.A336S variant (also known as c.1006G>T), located in coding exon 11 of the CDC73 gene, results from a G to T substitution at nucleotide position 1006. The alanine at codon 336 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
