NM_003738.5(PTCH2):c.3431G>A (p.Gly1144Glu) was classified as Uncertain significance for Basal cell nevus syndrome 1 by St. Jude Molecular Pathology, St. Jude Children's Research Hospital, citing St. Jude Assertion Criteria 2020. This variant lies in the PTCH2 gene (transcript NM_003738.5) at coding-DNA position 3431, where G is replaced by A; at the protein level this means replaces glycine at residue 1144 with glutamic acid — a missense variant. Submitter rationale: The PTCH2 c.3431G>A (p.Gly1144Glu) missense change has a maximum subpopulation frequency of 0.0017% in gnomAD v2.1.1 (PM2_supporting; https://gnomad.broadinstitute.org/variant/1-45288268-C-T). Five of six in silico tools predict a benign effect of this variant on protein function (BP4), but to our knowledge these predictions have not been confirmed by functional assays. To our knowledge, this variant has not been reported in individuals with Gorlin syndrome. In summary, this variant meets criteria to be classified as of uncertain significance based on the ACMG/AMP criteria: PM2_supporitng, BP4.
