Uncertain significance for Thrombocytopenia 2 — the classification assigned by St. Jude Molecular Pathology, St. Jude Children's Research Hospital to NM_014915.3(ANKRD26):c.3692C>G (p.Thr1231Ser), citing St. Jude Assertion Criteria 2020: The ANKRD26 c.3692C>G (p.Thr1231Ser) missense change has a maximum subpopulation frequency of 0.00088% in gnomAD v2.1.1 (PM2_Supporitng; https://gnomad.broadinstitute.org/variant/10-27322269-G-C ). This variant is not located in the 5’ UTR. Seven of seven in silico tools predict a benign effect of this variant on protein function (BP4), but these predictions have not been confirmed by functional studies. To our knowledge, this variant has not been reported in individuals with ANKRD26-related thrombocytopenia. In summary, this variant meets criteria to be classified as of uncertain significance based on the ACMG/AMP criteria: BP4.
