This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 3534, duplicating one base; at the protein level this means shifts the reading frame starting at glutamine residue 1179, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.3534dup variant is not present in publicly available population databases like 1000 Genomes, Exome Variant Server (EVS), Exome Aggregation Consortium (ExAC), Genome Aggregation Database (gnomAD), dbSNP and Indian Exome Database. The variant is also not present in our in-house exome database. The variant was not previously reported to ClinVar, Human Genome Mutation Database (HGMD) and OMIM databases in any affected individuals. In-silico pathogenicity prediction programs like MutationTaster2, CADD, Varsome etc. predicted this variant to be likely deleterious. The variant causes frameshift at the 1179th amino acid position that creates a premature stop codon at 1192th amino acid position of the altered transcripts that may either results into a truncated protein or causes nonsense mediated decay of the mRNA.
