Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_003680.4(YARS1):c.1034C>T (p.Ser345Leu), citing Ambry Variant Classification Scheme 2023: The p.S345L variant (also known as c.1034C>T), located in coding exon 9 of the YARS gene, results from a C to T substitution at nucleotide position 1034. The serine at codon 345 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
