This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 1064, where C is replaced by T; at the protein level this means replaces serine at residue 355 with leucine — a missense variant. Submitter rationale: The p.S355L variant (also known as c.1064C>T), located in coding exon 9 of the PTEN gene, results from a C to T substitution at nucleotide position 1064. The serine at codon 355 is replaced by leucine, an amino acid with dissimilar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955).
