NM_001365951.3(KIF1B):c.1130C>T (p.Thr377Ile) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T371I variant (also known as c.1112C>T), located in coding exon 11 of the KIF1B gene, results from a C to T substitution at nucleotide position 1112. The threonine at codon 371 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
