Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001110792.2(MECP2):c.1116_1117delinsTA (p.Pro373Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MECP2 c.1080_1081delinsTA (p.Pro361Thr) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 202050 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1080_1081delinsTA in individuals affected with Rett Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014, however a similar missense variant with the same amino acid change [NM_001110792.2:c.1117C>A|p.Pro373Thr] has been reported by two ClinVar submitters: both classified the variant as of uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
