Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001364171.2(ODAD1):c.1148T>C (p.Leu383Ser), citing Ambry Variant Classification Scheme 2023: The p.L346S variant (also known as c.1037T>C), located in coding exon 9 of the CCDC114 gene, results from a T to C substitution at nucleotide position 1037. The leucine at codon 346 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
