Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001364171.2(ODAD1):c.1157G>A (p.Arg386His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ODAD1 gene (transcript NM_001364171.2) at coding-DNA position 1157, where G is replaced by A; at the protein level this means replaces arginine at residue 386 with histidine — a missense variant. Submitter rationale: This sequence change replaces arginine with histidine at codon 349 of the CCDC114 protein (p.Arg349His). The arginine residue is weakly conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (rs200697095, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with CCDC114-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
