Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001349.4(DARS1):c.767T>A (p.Met256Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with lysine at codon 256 of the DARS protein (p.Met256Lys). The methionine residue is highly conserved and there is a moderate physicochemical difference between methionine and lysine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Met256 amino acid residue in DARS. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 23643384). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals affected with DARS-related conditions. This variant is not present in population databases (ExAC no frequency).
