Uncertain significance for Severe early-onset pulmonary alveolar proteinosis due to MARS deficiency; Charcot-Marie-Tooth disease axonal type 2U — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004990.4(MARS1):c.2701_*3del (p.Ter901del), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the MARS gene (p.*901Thrext*8). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last termination codon of the MARS protein and extend the protein by 8 additional amino acid residues. This variant is present in population databases (no rsID available, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with MARS-related conditions. ClinVar contains an entry for this variant (Variation ID: 1681809). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
