Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020702.5(MYORG):c.1069A>T (p.Thr357Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with serine at codon 357 of the KIAA1161 protein (p.Thr357Ser). The threonine residue is moderately conserved and there is a small physicochemical difference between threonine and serine. This variant is present in population databases (rs749542221, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with KIAA1161-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0").
